摘    要:In this talk, I present a mathematical framework to optimize combination immunotherapy for breast cancer, focusing on the interplay between mRNA-based cancer vaccines and anti-CTLA-4 monoclonal antibodies. While mRNA vaccines enhance T-cell activation and antitumor immunity, the CTLA-4 checkpoint pathway suppresses T-cell responses, limiting therapeutic efficacy. Anti-CTLA-4 antibodies can counteract this suppression but often induce dose-limiting immune-related toxicities. To address this challenge, we develop a mechanistic model that integrates immune-tumor dynamics, vaccine-induced priming, and checkpoint inhibition within the tumor microenvironment. Our simulations reveal that sequencing anti-CTLA-4 therapy after vaccination—and calibrating its dosage relative to vaccine strength—can maximize efficacy while minimizing toxicity. These findings provide a quantitative rationale for designing clinical trials and personalized immunotherapy regimens.
简    介:薛玲,哈尔滨工程大学数学科学学院教授、博士生导师、生物数学团队学科带头人、龙江学者特聘教授、黑龙江省专业技术领军人才梯队后备带头人、黑龙江省工程与应用重点实验室副主任。在美国堪萨斯州立大学获得博士学位后在美国杜兰大学数学系与数学计算科学中心做博士后,师从美国工业与应用数学会前主席James Mac Hyman教授;随后受到加拿大太平洋数学研究院和加拿大曼尼托巴大学数学系的联合资助,在加拿大曼尼托巴大学数学系合作交流。在Journal of Mathematical Biology 、SIAM J Appl Math、Bulletin of Mathematical Biology等SCI 期刊发表论文50余篇,担任T2期刊Bulletin of Mathematical Biology、PloS Computational Biology 等的编委。
